My FireWire or USB drive doesn't show up in the pop-up.

Most FireWire and nearly all USB drives are shipped formatted as FAT32 (Windows/DOS) volumes, rather than as HFS+ (Mac OS Extended) volumes. Since you can't create a bootable clone on a FAT32 volume, SuperDuper! doesn't display those volumes in the destination pop-up.

To use this kind of drive, simply use Disk Utility to erase it, using "Mac OS Extended (Journaled)" as the format. Once erased and formatted, SuperDuper! will allow the drive to be used.
