Cannot unmount primary laptop drive after booting from SuperDuper copy

Hi all,

I have used SD to clone my hard disk on my MB Pro and booted from that image successfully. I now want to unmount the laptop's drive so I can perform a full defrag using iDefrag, however when I try to do this, I get 'The disk "Macintosh HD" could not be unmounted. Make sure that all applications and files are closed on this disk.'

I have nothing running at all so I guess there's an internal process keeping a file open. Can anyone suggest what action I might take to unmount this drive?

I am using a MB Pro running 10.4.10.
