iTunes Library/Music in Backup

If I backup a folder with the iTunes Library folders in it, does it matter if iTunes is running with it's library loaded during the backup? Obviously if I imported a file into iTunes during the backup, it may or may not be backed up depending on where the backup process is. I am more concerned that files will be locked or corrupted and not viable if they are restored.

Not generally, no, but since the iTunes database is a (large) file that can get updated, you'll want to ensure you're not "actively" using iTunes while you back it up.
