Copied volume in Finder - can I now Smart Update?

I've copied a volume from one external drive to another using Finder. Can I now use SuperDuper's Smart Update or Copy Newer to keep the two volumes the same - or will it only work if I've done the initial copy with SuperDuper? They're backups of media files - not startup volumes.
