Super duper fails to back-up after 50sec.
 

Unable to back-up.
Keep getting "Failed to copy to Macintosh HD" error.
Is anyone able to help?

Your source drive has some damage on it that needs to be fixed; see the "Troubleshooting" section of the User's Guide for the steps to take.

Fixed the problem by uninstalling all lexmark printer files and reinstalling them. Clue was in the log.

Yes, that'll do it too, but I strongly advise repairing the drive as indicated in Troubleshooting, since the files shouldn't have been damaged in the first place...
