I would like to configure SuperDuper! to automatically do the following before making a duplicate and reverse it afterwards:

- Turn TimeMachine off
- Turn Wi-Fi off
- Quit Mail and specific other applications.

I've tried to find out how in SuperDuper's help and documentation but can't. It must also surely be possible with Automator but I haven't figured that out either.

Is there a way?

You can sort of use a before-copy shell script, but it requires knowing how to use shell scripting. You generally wouldn't need to do these things, though - why are you doing so?
